
KiraGen completed its first exploratory in vivo proof-of-concept study for a multiplex-engineered CAR-T precursor on December 2, 2024.
In a subcutaneous GBM xenograft model, a five-edit precursor demonstrated improved tumor control relative to unedited and single-edit controls under the tested conditions. The study provided early in vivo support for KiraGen's central thesis: CAR-T cells may need simultaneous protection from several suppressive pathways to remain active in solid tumors.
This work established an initial biological foundation for the company's multiplex-engineering strategy and informed subsequent candidate development. The study was exploratory and preclinical, and it did not evaluate an orthotopic or patient-derived model. Related findings were later presented during a GenScript exhibitor showcase at ASGCT 2025.
